Overview
Tesamorelin (TH9507) is a synthetic analogue of human growth hormone-releasing hormone (GHRH) comprising the full 44-amino-acid sequence of endogenous GHRH, modified at the N-terminus with a trans-3-hexenoic acid moiety to confer resistance to DPP-IV-mediated cleavage. This modification extends its plasma half-life while preserving full binding affinity at the pituitary GHRH receptor.
Unlike truncated GHRH analogues, tesamorelin engages the receptor with the native full-length peptide configuration, producing robust, sustained GH secretion with downstream elevation of IGF-1. It is one of the most extensively characterized synthetic GHRH analogues in clinical research, with a substantial body of controlled trial data examining its effects on body composition — particularly visceral adipose tissue.

Clinical trials have documented tesamorelin's ability to significantly reduce visceral adipose tissue (VAT) — the metabolically active fat depot surrounding abdominal organs — as well as hepatic fat content and liver-associated inflammatory markers.
