Overview
SS-31 (Elamipretide / MTP-131 / Bendavia) is a synthetic tetrapeptide with the sequence D-Arg–Tyr(2,6-diMe)–Lys–Phe, designed to selectively concentrate at the inner mitochondrial membrane. Its cationic and aromatic residues enable it to interact with cardiolipin, a phospholipid unique to the inner mitochondrial membrane that is essential for the structural integrity and function of the mitochondrial electron transport chain.
By stabilizing the cardiolipin–cytochrome c interaction and protecting cristae architecture, SS-31 is hypothesized to maintain efficient electron transport, reduce electron leak and reactive oxygen species (ROS) generation, and preserve ATP synthesis capacity under conditions of mitochondrial stress or damage.
Research Highlights
Preclinical studies have demonstrated SS-31's protective effects in ischemia-reperfusion injury models across cardiac, renal, skeletal muscle, and neuronal tissue. In these models, it has been shown to reduce oxidative damage, preserve mitochondrial membrane potential, improve ATP output, and attenuate post-ischemic organ dysfunction.
Research has also explored SS-31 in the context of aging-related mitochondrial decline, muscular dystrophy, metabolic syndrome, and Alzheimer's disease models. Its ability to selectively target the inner mitochondrial membrane without accumulating to toxic concentrations — unlike some other mitochondria-targeting cations — is a mechanistically important property that has fueled its use as a research probe for mitochondrial biology and therapeutic development.
