Overview

GHK-Cu (Glycyl-L-histidyl-L-lysine copper complex) is a naturally occurring tripeptide found in human plasma, saliva, and urine that forms a high-affinity complex with copper(II) ions. Plasma concentrations of GHK-Cu decline significantly with age, a pattern that has driven research interest in its potential role in age-associated tissue deterioration and wound healing capacity.

As a copper chaperone peptide, GHK-Cu modulates the activity of numerous copper-dependent enzymes involved in extracellular matrix remodeling, including lysyl oxidase, which crosslinks collagen and elastin. This activity underlies its studied roles in wound repair, skin tissue organization, and anti-fibrotic signaling.

Research Highlights

Preclinical studies have documented GHK-Cu's ability to stimulate collagen, elastin, and glycosaminoglycan synthesis in fibroblasts, accelerate wound healing across skin, tendon, and bone tissue models, promote hair follicle proliferation, and exert antioxidant effects through free radical neutralization and superoxide dismutase upregulation.

Gene expression research has revealed that GHK-Cu modulates a surprisingly broad set of biological pathways — including genes related to inflammation, DNA repair, anti-tumor defense, and metabolic function — suggesting it functions as a pleiotropic signaling molecule rather than a simple tissue-repair agent. Its anti-inflammatory effects, including downregulation of TNF-alpha and other pro-inflammatory cytokines, are also documented in multiple preclinical models.
